چکیده

Rifampicin, one of the primary tuberculosis drugs, is a PGP substrate that is encoded by MDR 1 gene. The objectives of the research was to study the impacts of MDR1 gene polymorphism toward plasma rifampicin levels in Javanese patients with pulmonary tuberculosis. The subjects of this research are Javanese tuberculosis patients treated by fixed dosage combination (FDC) of anti-tuberculosis drugs containing rifampicin. There were 74 patients. The examination of MDR1 was conducted by employing restriction fragment length polymorphism (RFLP-PCR) method. Blood samplings to assays the rifampicin level were done on day 1; 4; 28; and 56. The measurement of rifampicin levels was done by high performance liquid chromatography (HPLC) method. There are significant difference on the level of plasma rifampicin between MDR1 CC, CT and TT with p <0,05. The polymorphism of MDR1 TT gene increase plasma rifampicin levels in Javanese patients with pulmonary tuberculosis.

Mutations in the RpoB Gene of Multidrug-Resistant Mycobacterium Tuberculosis Isolates from Semarang, Indonesia

ABSTRACTTuberculosis is an infectious disease which occurs widely in the world and becomes one of the top 10 causes of death worldwide. Mutations of the RpoB gene cause the resistance of Mycobacterium tuberculosis to rifampicin that contributes to the occurrence of MDR-TB. This study aimed to determine the pattern of rpoB gene polymorphisms in the MDR M. tuberculosis in Semarang, Indonesia. Mos...

Association of Acute Lymphoblastic Leukemia and MDR1 Gene Polymorphism in an Ethnic Iranian Population

Background: The frequency of the multi-drug resistance 1 (MDR1) gene C3435T polymorphism differs in various ethnical populations such as Asian, African, and Caucasians populations. A silent C3435T polymorphism in exon 26 of MDR1 has been reported to be associated with a decreased expression of P-gp in TT genotypes carriers compared with CC genotypes carriers. Materials and Methods: To evaluate ...

The NRAMP1 polymorphism as a risk factor for tuberculous spondylitis

UNLABELLED In the present study, we analysed the association between the incidence of tuberculous spondylitis with the Natural Resistance Associated Macrophage Protein 1 (NRAMP1, also known as Solute Carrier Family 11a member1) polymorphism by studying the genetic segregation of this polymorphism and the incidence of the disease among members of the West Javanese population undergoing surgery f...

بررسی فراوانی پلی مورفیسم ژنتیکی C3435T ژن MDR1 در یک جمعیت سالم استان مازندران

Background and purpose: The human multidrug resistance gene (MDR1) encodes for P-glycoprotein (P-gp) which is a transmembrane transporter protein acts as an efflux pump for a number of xenobiotics. It plays a protective role for cells against DNA damage caused by toxins and drugs. The wobble C3435T polymorphism at exon 26 has been associated with different expression levels and activities of th...

Lack of Association of SP110 Gene Polymorphisms with Pulmonary Tuberculosis in Golestan Province, Iran

ABSTRACT Background and Objectives: Mycobacterium tuberculosis is the causative agent of pulmonary tuberculosis, a main public health problem that results in 1.5 million deaths annually. A number of epidemiological studies suggested that host genetic factors could play a main role in susceptibility to tuberculosis infection. SP110 is an i...
